Chagas Disease: Chronic Chagas Cardiomyopathy

Natalia Giraldo Echavarria et al.

Summary: Chagas disease is a tropical vector-borne infection caused by the parasite Trypanosoma cruzi, endemic in South and Central America but increasingly becoming a burden in the United States and Europe due to human migration. Most patients are asymptomatic during the acute phase, with around 30% progressing to detectable organ damage mainly affecting the cardiovascular and digestive systems in the chronic symptomatic phase, which can lead to Chagas cardiomyopathy, a leading cause of nonischemic cardiomyopathy in Latin America. Treatment during the parasitic infection is possible, but efficacy during the advanced chronic symptomatic phase remains uncertain. Further studies are needed to improve clinical outcomes and refine treatment recommendations.

Determinism and contingencies shaped the evolution of mitochondrial protein import

Samuel Rout et al.

Summary: The study reveals that the mitochondrial import receptors ATOM46 and ATOM69 of Trypanosoma brucei have different substrate preferences, with ATOM46 favoring hydrophilic proteins and ATOM69 preferring hydrophobic substrates. While ATOM46 mainly uses electrostatic interactions for substrate binding, ATOM69 primarily uses hydrophobic interactions.

Novel Cytoskeleton-Associated Proteins in Trypanosoma brucei Are Essential for Cell Morphogenesis and Cytokinesis

Marina Schock et al.

Summary: Trypanosome brucei, the causative agent of African sleeping sickness, has a highly ordered subpellicular microtubule cytoskeleton associated with various proteins regulating morphology, motility, and virulence. Identified protein CAP50 colocalises with microtubules but not with the flagellum, playing a crucial role in maintaining cellular integrity. Depletion of CAP50, along with CAP52 and CAP42, results in defects in cytokinesis, morphology, and microtubule organization.

Targeting the mitochondrial chaperone TRAP1: strategies and therapeutic perspectives

Stefano A. Serapian et al.

Summary: TRAP1, the mitochondrial isoform of heat shock protein (Hsp)90 chaperones, plays a crucial role in regulating metabolism and organelle homeostasis in various pathological states. Developing TRAP1-directed strategies, including lead modification with mitochondria delivery groups and computational discovery of allosteric sites and ligands, opens up unique opportunities in addressing fundamental questions on its biology and unveiling new therapeutic approaches. Predicting the activities of TRAP1-selective allosteric ligands and optimizing target engagement to avoid side effects are crucial in this endeavor.

The Bacterial Hsp90 Chaperone: Cellular Functions and Mechanism of Action

Sue Wickner et al.

Summary: Heat shock protein 90 (Hsp90) is a molecular chaperone that folds and remodels proteins, regulating the activity of substrate proteins. Bacterial Hsp90 collaborates with Hsp70 and its cochaperones for protein remodeling, while eukaryotic Hsp90 involves over 20 cochaperones in addition to Hsp70. Further exploration of the bacterial chaperone system may provide insights into the complex eukaryotic Hsp90 system.

The Mitochondrial Hsp90 TRAP1 and Alzheimer's Disease

Francoise A. Dekker et al.

Summary: This article discusses the molecular mechanism and cellular function of TRAP1, and explores possible links to Alzheimer's Disease (AD). TRAP1, as a member of the Hsp90 family, plays a crucial role in chaperoning essential cellular proteins despite not being regulated by co-chaperones like its cytosolic counterparts. It is also involved in maintaining mitochondrial integrity by regulating mitochondrial pore opening through Cyclophilin D.
