Does a Hypertrophying Muscle Fibre Reprogramme its Metabolism Similar to a Cancer Cell?

Henning Wackerhage et al.

Summary: There are metabolic differences between growing and non-growing tissues, and hypertrophying muscle reprograms its metabolism similar to cancer cells. Increased glucose uptake in hypertrophying muscle may explain the negative association with type 2 diabetes mellitus and obesity.

A glitch in the matrix: the pivotal role for extracellular matrix remodeling during muscle hypertrophy

Camille R. Brightwell et al.

Summary: Multinuclear muscle fibers are the largest cells in skeletal muscle and play a crucial role in muscle growth. However, mononuclear cells in the extracellular matrix (ECM) also contribute to muscle hypertrophy. Proper remodeling and maintenance of the ECM is essential for an appropriate response to resistance training in adult muscles. The ECM remodeling process involves various cells and signals, and the acellular collagen-rich ECM acts as a mechanical signal transducer and repository of growth factors that influence muscle fiber hypertrophy. This review highlights the impact of the ECM and its cellular components on loading-induced muscle hypertrophy.

The titin N2B and N2A regions: biomechanical and metabolic signaling hubs in cross-striated muscles

Robbert J. van der Pijl et al.

Summary: This review highlights the biological roles and properties of the titin N2B and N2A regions in health and disease, with a special emphasis on the functions of Ankrd and FHL proteins as mechanosensors modulating muscle-specific signaling and muscle growth. The sarcomere region also emerged as a hotspot for the modulation of passive muscle mechanics through altered titin phosphorylation and splicing, as well as tethering mechanisms linking titin to the thin filament system.

Genetic and epigenetic regulation of skeletal muscle ribosome biogenesis with exercise

Vandre C. Figueiredo et al.

Summary: The study reveals the genetic and rDNA gene-wide epigenetic regulation of ribosome biogenesis in response to exercise, highlighting the roles of rDNA dosage and CpG methylation in skeletal muscle. This indicates a genetic predisposition for hypertrophic responsiveness and preferential up-regulation of ribosome biogenesis with resistance exercise. The findings expand our understanding of the genetic and epigenetic factors involved in exercise-induced changes in ribosome biogenesis.

Evidence of myomiR regulation of the pentose phosphate pathway during mechanical load-induced hypertrophy

Taylor Valentino et al.

Summary: The research identifies the molecular and cellular mechanisms regulating skeletal muscle hypertrophy using the rodent synergist ablation model, and examines the metabolic pathways in muscle during mechanical overload (MOV) through gene expression analysis. The study suggests that enhanced redox metabolism via NADPH production may play a key role in muscle hypertrophy induced by MOV, potentially regulated by a myomiR network.

Muscle ankyrin repeat protein 1 (MARP1) locks titin to the sarcomeric thin filament and is a passive force regulator

Robbert J. van der Pijl et al.

Summary: This study demonstrates that MARP1 interacts with F-actin and forms a complex with titin-N2A, which leads to the locking of titin to the thin filament, resulting in increased passive force. The mechanism proposed here suggests that in stressed muscle, MARP1 plays a role in protecting the sarcomere from mechanical damage.
