The SARS-CoV-2 nucleocapsid phosphoprotein forms mutually exclusive condensates with RNA and the membrane-associated M protein

Shan Lu et al.

Summary: The SARS-CoV-2 nucleocapsid (N) protein binds viral RNA and undergoes liquid-liquid phase separation driven by its central intrinsically-disordered region, modulated by phosphorylation. Additionally, the N protein interacts with the membrane protein M to form mutually exclusive compartments in a three-component mixture with RNA.

A novel cell culture system modeling the SARS-CoV-2 life cycle

Xiaohui Ju et al.

Summary: A BSL-2 cell culture system for production of replication-competent SARS-CoV-2 virus-like-particles was developed. This system allows for the complete viral life cycle to be achieved and enhances biosafety measures. High-throughput antiviral screening was conducted, leading to the discovery of new drugs against SARS-CoV-2 infection.

Structural and functional consequences of NEDD8 phosphorylation

Katrin Stuber et al.

Summary: The study reveals that NEDD8 can be phosphorylated at S65, affecting its structural dynamics similar to Ub. Phosphorylated NEDD8 has a distinct interactome compared to unmodified Ub and NEDD8, and it can more effectively regulate HSP70 protein activity.

1H, 13C and 15N Backbone chemical shift assignments of the n-terminal and central intrinsically disordered domains of SARS-CoV-2 nucleoprotein

Serafima Guseva et al.

Summary: The nucleoprotein (N) from SARS-CoV-2 plays an important role in the viral replication transcription complex and in protecting the viral genome from the host immune system. N is a 419 amino acid multidomain protein with two folded domains and a predicted intrinsically disordered region, for which atomic resolution information is currently lacking.

Multivalent binding of the partially disordered SARS-CoV-2 nucleocapsid phosphoprotein dimer to RNA

Heather M. Forsythe et al.

Summary: The nucleocapsid phosphoprotein N plays crucial roles in various processes of the SARS-CoV-2 infection cycle by interacting with RNA and forming large oligomers. Biophysical and biochemical studies reveal that N protein is partially disordered, consisting of an RNA-binding domain and a dimerization domain, and can assemble as tight dimers with transient interactions. The study also identifies a heterogeneous mixture of N-RNA complexes, highlighting the importance of N's bivalent structure in binding to multivalent sites on viral RNA.
