Reprogramming cholesterol metabolism in macrophages and its role in host defense against cholesterol-dependent cytolysins

Min-Sub Lee et al.

Summary: Cholesterol plays a critical role in maintaining the integrity, fluidity, and biochemical function of mammalian cells. Macrophages can rapidly reprogram their cholesterol metabolism in response to immune activation signals. This review discusses current knowledge of cellular cholesterol homeostasis and highlights the reprogramming of cholesterol metabolism in macrophages during immune responses. It also explores the effects of these changes on sensitivity to microbial toxins and the potential therapeutic applications in diseases associated with tissue damage caused by cholesterol-dependent toxins.

Rapid GPR183-mediated recruitment of eosinophils to the lung after Mycobacterium tuberculosis infection

Andrea C. Bohrer et al.

Summary: The influx of eosinophils into the lungs is associated with various immune responses, including allergies and infections. This study reveals that eosinophils also accumulate in lung lesions during inflammation caused by Mycobacterium tuberculosis (Mtb) infection. These cells migrate into the lungs as early as one week after Mtb exposure and their recruitment is dependent on the oxysterol receptor GPR183. Eosinophils directly interact with bacilli-laden alveolar macrophages, indicating their role in sensing and responding to Mtb infection.

Clearance of apoptotic cells by lung alveolar macrophages prevents development of house dust mite-induced asthmatic lung inflammation

Haruka Miki et al.

Summary: The study showed that engulfment of apoptotic cells can suppress allergic asthma by promoting the production of regulatory T cell-inducing molecules, inhibiting inflammatory cytokine production, and increasing the sensitivity of lung alveolar macrophages to adenosine signals. The findings suggest that targeting lung macrophages to enhance their phagocytic capacity and regulatory functions could be a potential strategy for suppressing allergic responses.

Corticosteroid insensitivity persists in the absence of STAT1 signaling in severe allergic airway inflammation

Brandon W. Lewis et al.

Summary: The study exposed cultured murine neurons and astrocytes to six prescription medications that cross the placenta and blood-brain barriers, finding that these drugs affected cholesterol biosynthesis, resulting in an increase in sterol intermediates and a decrease in cholesterol levels in the cells. Four prescription medications primarily acted on the DHCR7 enzyme, leading to an increase in 7-dehydrocholesterol levels in neurons and astrocytes.

LXR directly regulates glycosphingolipid synthesis and affects human CD4+T cell function

Kirsty E. Waddington et al.

Summary: The liver X receptor (LXR) plays a key role in regulating cholesterol, fatty acid, and phospholipid metabolism in T cells. Activation of LXR alters the balance of glycosphingolipids and cholesterol in the plasma membrane, resulting in decreased lipid order and dampening proinflammatory T cell function. This study highlights the importance of LXR in modulating T cell immune signaling and function through lipid metabolism regulation.
