PBK Model-Based Prediction of Intestinal Microbial and Host Metabolism of Zearalenone and Consequences for its Estrogenicity

Summary: This study developed physiologically-based kinetic models for rat and human, incorporating intestinal microbial and hepatic metabolism of zearalenone (ZEN) to predict systemic concentrations of ZEN. The results showed that hepatic metabolism primarily drives the formation of the estrogenic metabolite alpha-zearalenol (alpha-ZEL). The models predicted that internal concentrations of ZEN and alpha-ZEL are significantly below levels known to induce estrogenicity in vitro at the tolerable daily intake.
