Engineering Saccharomyces cerevisiae for Hyperproduction of β-Amyrin by Mitigating the Inhibition Effect of Squalene on β-Amyrin Synthase

Summary: The study aims to enhance the production of beta-amyrin in Saccharomyces cerevisiae through peroxisome compartmentalization and the push-pull-restrain strategy. The results show that peroxisome compartmentalization can mitigate the inhibitory effect of overaccumulated squalene on beta-amyrin synthase activity and significantly increase its production.

Supplementation of this oil resulted in changes in lipid profiling and activation of pathways related to memory and cognition improvement. Summary: This study developed a novel method to increase the supply of acetyl-CoA in Saccharomyces cerevisiae and improve the production of downstream products. By optimizing metabolic pathways and conducting genetic engineering, the production of mevalonic acid was successfully increased, and further improvement was achieved by dynamically downregulating gene expression. The final strain produced a high yield of lycopene.

Summary: Adrenoleukodystrophy (ALD) is an inherited disorder characterized by accumulation of very long-chain fatty acids. We investigated nervonic acid as a potential therapy for ALD and found that it can reverse lipid accumulation and protect cells from oxidative damage. This suggests that nervonic acid could be a therapeutic option for ALD.
