Enhancement of the Isomerization Activity and Thermostability of Cellobiose 2-Epimerase from Caldicellulosiruptor saccharolyticus by Exchange of a Flexible Loop

Lu Wang et al.

Summary: This study aimed to optimize the catalytic performances of CEs by flexible loop exchange, resulting in mutants with improved isomerization activity and thermostability. Molecular dynamics simulations showed that loop exchange enlarged the entrance of the active site for substrate binding and benefited proton transfer. This work demonstrated the importance of the flexible loop surrounding the active site in determining the isomerization activity and thermostability of CEs.

Flexible Loop in Carbohydrate-Binding Module 48 Allosterically Modulates Substrate Binding of the 1,4-α-Glucan Branching Enzyme

Haimin Jiang et al.

Summary: The study focused on the crystal structure analysis of the GBE enzyme, and found that the CBM48 domain plays a crucial role in substrate binding and enzymatic activity of RoGBE. Comparison of RoGBE with other GBEs revealed that CBM48 of RoGBE has a longer flexible loop, and truncation of this loop resulted in reduced enzyme activity.

Microbial starch debranching enzymes: Developments and applications

Wei Xia et al.

Summary: Starch debranching enzymes (SDBEs) play a crucial role in hydrolyzing alpha-1,6 glycosidic bonds in polysaccharides and are essential for various industrial processes. While the substrate specificities of pullulanases and isoamylases differ, both enzymes belong to glycoside hydrolase families and have similarities in their core catalytic domains. Efforts have been made to improve the catalytic efficiency, thermostability, and expression level of SDBEs through enzyme engineering and fermentation strategies. Researchers have also summarized the diverse biochemical properties and features of pullulanase and isoamylase from different sources, aiming to broaden the applications of SDBEs in various industries.
