Emerging role of ferroptosis in breast cancer: New dawn for overcoming tumor progression

Shiyao Sui et al.

Summary: Breast cancer is a significant threat to women's health, and ferroptosis may offer a new direction for breast cancer treatment. However, the complete appearance of ferroptosis in breast cancer and its potential combination with traditional anti-breast cancer drugs remain uncertain. This review explores the roles of various factors associated with ferroptosis in breast cancer, provides evidence of ferroptosis induction by traditional drugs and elimination of breast cancer cells by ferroptosis inducers, and discusses the prospects and potential obstacles of using ferroptosis inducers in breast cancer therapy.

Cancer stem cell regulated phenotypic plasticity protects metastasized cancer cells from ferroptosis

Mingming Wu et al.

Summary: The study reveals the role of breast cancer stem cell (BCSC) secretome in regulating the stem cell population, tumor-initiating capacity, and metastasis. DKK1 is identified as a key factor secreted by BCSCs that promotes differentiation and metastatic outgrowth. DKK1 increases the expression of SLC7A11 to protect metastasizing cancer cells from ferroptosis. Combination therapy with a ferroptosis inducer and a DKK1 inhibitor shows synergistic effects in reducing metastasis.

Comprehensive Analysis of a Ferroptosis Pattern and Associated Prognostic Signature in Acute Myeloid Leukemia

Zelong Cui et al.

Summary: Ferroptosis is a form of programmed cell death that is widespread. This study investigates the association between ferroptosis and AML outcome using ferroptosis-related genes. Two subtypes of AML were identified and linked to immunological abnormalities, mutation landscape, pathway dysregulation, and clinical outcome. A 13-gene prognostic model was developed and validated in independent AML cohorts and pan-cancer cohorts. The model's translational value was enhanced by its ability to discriminate the sensitivity of chemotherapeutic drugs and ferroptosis inducers. The study sheds new light on the relationship between ferroptosis and AML, and has potential implications for clinical management.

Construction and Validation of a 6-Ferroptosis Related Gene Signature for Prognosis and Immune Landscape Prediction in Melanoma

Zhanghui Yue et al.

Summary: In this study, a 6-gene signature related to ferroptosis was constructed to predict the prognosis of melanoma patients. Patients in the high-risk group had a worse prognosis and lower immune function. Additionally, the low-risk group showed higher expression of immune checkpoint molecules and greater sensitivity to immunotherapy. Some small molecular drugs were also identified as potential treatments for melanoma.

Extracellular HMGB1 blockade inhibits tumor growth through profoundly remodeling immune microenvironment and enhances checkpoint inhibitor-based immunotherapy

Pascale Hubert et al.

Summary: In this study, it was demonstrated that targeting extracellular HMGB1 can significantly remodel the tumor immune microenvironment for more effective cancer therapy. Inhibition of HMGB1 led to reductions in immunosuppressive cells, such as myeloid-derived suppressor cells and regulatory T lymphocytes, while also enhancing the activation of immune cells like dendritic cells. Additionally, blocking HMGB1 improved the efficacy of anti-PD-1 cancer immunotherapy, suggesting that extracellular HMGB1 blockade could enhance current cancer immunotherapies.
