DNA-Lysozyme Nanoarchitectonics: Quantitative Investigation on Charge Inversion and Compaction

Summary: The interaction between DNA and proteins is important for both basic research in biology and potential applications in nanotechnology. This study investigated the complexes formed by lambda DNA and lysozyme in a dilute aqueous solution. It was found that lysozyme effectively neutralizes the positive charge of DNA, inducing charge inversion. The compact and positively charged structure of lysozyme contributes to its high efficiency in charge neutralization. Increasing the concentration of lysozyme leads to changes in both DNA mobility and condensing force, as well as the morphology of the DNA-lysozyme complex.

Evaluation of weak interactions of proteins and organic cations with DNA duplex structures

Summary: This study investigates the effects of cellular crowding on nucleic acid structures and finds that basic globular proteins can stabilize long loop structures but not fully matched duplexes. The effects of basic proteins on stability differ from those of polypeptides and polyamines. These findings provide new insights into the interaction of organic cations with nucleic acid structures.

Impact of denaturing agents on surface properties of myoglobin solutions

Summary: The addition of denaturants strongly affects the surface properties of aqueous myoglobin solutions, leading to different effects compared to mixed solutions with other globular proteins. The dynamic surface elasticity of myoglobin solutions with GuHCl shows at least two adsorption steps at denaturant concentrations higher than 1 M, a rapidly increasing elasticity followed by a slower growth to abnormally high values. Protein aggregation in the surface layer may contribute to the higher surface elasticity in myoglobin/GuHCl solutions, with evidence supported by ellipsometry and Brewster angle microscopy. Surfactant addition to protein solutions results in the formation of myoglobin/surfactant complexes, displaying multi-step adsorption kinetics different from solutions of other globular proteins mixed with ionic surfactants. Ellipsometry and infrared reflection-absorption spectroscopy allow tracking the adsorption and displacement of the complexes at high surfactant concentrations.

DNA Interaction with a Polyelectrolyte Monolayer at Solution-Air Interface

Summary: This study reveals that a novel approach can confine specific polyelectrolyte at the solution-air interface with elevated ionic strength, leading to non-monotonic kinetic dependencies of surface properties in the formation of mixed PDAHMAC/DNA aggregates. Despite different aggregation mechanisms, the micromorphology of the mixed PDAHMAC/DNA does not noticeably depend on the initial surface pressure.
