Summary: This study demonstrated the involvement of ferroptosis in retinal ischemia reperfusion (IR) in mice, in addition to necroptosis and apoptosis. The inhibitor Ferrostatin-1 showed the best therapeutic effect in protecting retinal ganglion cells (RGCs) from IR. Inhibition of activated microglia, Muller cells, immune responses, and intracellular reactive oxygen species accumulation was observed after treatment with each inhibitor. Combination therapy with Ferrostatin-1, z-VAD-FMK, and Necrostatin-1 was more effective in preventing IR-induced RGC death than using any inhibitor alone.

Inflammation in Glaucoma: From the back to the front of the eye, and beyond

Summary: The pathophysiology of glaucoma is complex and multifactorial, involving elevated intraocular pressure and impaired retinal blood flow as primary factors. Oxidative stress, cell senescence, mitochondrial dysfunction, excitotoxicity, and neuroinflammation play crucial roles in glaucoma progression. Inflammatory processes in various ocular structures from the posterior to the anterior segment are present in glaucoma, supporting the hypothesis of dysregulation towards a pro-inflammatory phenotype. Possible anti-inflammatory therapeutic approaches for glaucoma are also discussed.
