Renal gluconeogenesis: an underestimated role of the kidney in systemic glucose metabolism

The kidney contributes significantly to systemic gluconeogenesis, with lactate being the main substrate. Impaired renal gluconeogenesis in acute and chronic kidney disease can lead to systemic metabolic disturbances. This review highlights the importance of kidney gluconeogenesis and its implications in kidney disease.

Exploration of Novel Hepatitis B Virus Capsid Assembly Modulators by Integrated Molecular Simulations

The impact of diabetes and osteoarthritis on the occurrence of stroke, acute myocardial infarction, and heart failure among older adults with non-valvular atrial fibrillation in Hawaii: a retrospective observational cohort study

Summary: This study aimed to examine the risks for stroke, acute myocardial infarction (AMI), and heart failure (HF) among older adults with non-valvular atrial fibrillation in Hawaii by their diabetes and osteoarthritis status. The results showed that individuals with diabetes and osteoarthritis had higher risks for heart failure compared to those without either condition, while individuals with diabetes had higher risks for all three cardiovascular diseases. Variation in cardiovascular disease risks exists depending on the presence of diabetes and osteoarthritis among older adults with non-valvular atrial fibrillation in Hawaii.

Current and emerging gluconeogenesis inhibitors for the treatment of Type 2 diabetes

Summary: In recent decades, significant progress has been made in elucidating the pathways of gluconeogenesis, with many potential therapeutic targets identified. Lipid metabolism and hepatic lipogenesis have been found to play a major role in gluconeogenesis and insulin resistance. Future research will focus on the interplay between gluconeogenesis and adipose tissue metabolism, with an acceleration in the exploration of therapeutic RNA agents to inhibit gluconeogenesis. The balance between clinical benefit and adverse effects will determine the future development of new gluconeogenesis inhibitors.

Novel 2-substituted 6-oxo-1,6-dihydropyrimidine-5-carboxylic acids (ODCs) have been reported as XO inhibitors (XOIs) with remarkable activities. Through 3D-QSAR, docking, pharmacophore modeling, and MD studies, key interactions between ODCs and XO protein were identified, leading to the discovery of potential hit compounds for the development of novel XOIs.

It was also highlighted that diabetes time management has the potential to improve patient outcomes in individuals with T2DM. This study is the first to measure the influence of diabetes time management on diabetes self-care and to explore the relationship between diabetes time management and diabetes distress.
