FBXO32 targets PHPT1 for ubiquitination to regulate the growth of EGFR mutant lung cancer

Summary: Our study demonstrates that elevated expression of PHPT1 is associated with poor overall survival in lung cancer patients, particularly in those carrying EGFR mutations. PHPT1 plays a critical role in promoting the proliferation, clonogenicity, and growth of EGFR mutant lung cancer cells by activating the ERK/MAPK pathway. In addition, we identify FBXO32 as an E3 ubiquitin ligase for PHPT1, and its knockdown leads to PHPT1 accumulation and activation of the ERK/MAPK pathway, promoting lung cancer cell growth. These findings suggest that PHPT1 may serve as a potential biomarker and therapeutic target for acquired erlotinib resistance in lung cancer patients.
