CDNF and MANF regulate ER stress in a tissue-specific manner

Emmi Pakarinen et al.

Summary: Cerebral dopamine neurotrophic factor (CDNF) and mesencephalic astrocyte-derived neurotrophic factor (MANF) both have cytoprotective effects in neurodegenerative diseases. They belong to the same protein family and function as endoplasmic reticulum stress regulators. CDNF and MANF display functional redundancy in muscle but not in other tissues, regulating unfolded protein response (UPR) in a tissue-specific manner.

Cerebral dopamine neurotrophic factor protects and repairs dopamine neurons by novel mechanism

Paivi Lindholm et al.

Summary: Parkinson's disease is a progressive neurodegenerative movement disorder with currently no cure to stop dopaminergic decline or restore function of injured neurons. CDNF is an unconventional neurotrophic protein that has shown in animal models to protect and restore dopamine neurons, with promising safety and tolerability results in clinical trials. Further research is needed to validate its effectiveness as a potential therapeutic agent for neurodegenerative diseases like PD.

The cytoprotective protein MANF promotes neuronal survival independently from its role as a GRP78 cofactor

Ave Eesmaa et al.

Summary: MANF is a UPR regulator that promotes the survival of ER-stressed neurons, affecting multiple UPR pathways simultaneously. Its interaction with GRP78 as a cofactor is dispensable for its survival-promoting activity in neurons, suggesting a divergent role in UPR regulation and antiapoptotic effects.

Exploration of the Hsa-miR-1587-Protein Interaction and the Inhibition to CASK

Lulu Zhang et al.

Summary: Hsa-miR-1587 is capable of forming G-quadruplex structures and is overexpressed in multiple cancer cell lines. Through interactions with proteins like CASK, it affects the expression levels of downstream proteins reelin and p21, potentially involved in intracellular metabolic and transcriptional physiological processes. This study provides a new strategy for regulating the function of G-rich microRNAs.

New Cardiomyokine Reduces Myocardial Ischemia/Reperfusion Injury by PI3K-AKT Pathway Via a Putative KDEL-Receptor Binding

Leonardo Maciel et al.

Summary: CDNF is proposed as a new cardiomyokine that induces cardioprotection via KDEL receptor binding and PI3K/AKT activation. It has been shown to protect cardiomyocytes against endoplasmic reticulum stress and reduce ischemia/reperfusion injury in both in vitro and in vivo models.

Quantitative FRET (qFRET) Technology for the Determination of Protein-Protein Interaction Affinity in Solution

Jiayu Liao et al.

Summary: Protein-protein interactions are vital in life, and FRET is widely used to detect molecular interactions with high sensitivity and efficiency. Attempts to develop FRET into a quantitative measurement for interaction affinity have faced challenges, but the method shows great potential for studying difficult-to-express proteins and interactions in living cells.

Cerebral dopamine neurotrophic factor reduces α-synuclein aggregation and propagation and alleviates behavioral alterations in vivo

Katrina Albert et al.

Summary: CDNF interacts with alpha-synuclein to reduce its aggregation and internalization, while inducing the formation of insoluble phosphorylated alpha-synuclein. However, the beneficial effects of CDNF on rodent behavior do not seem to be related to the number of phosphorylated alpha-synuclein inclusions formed. Further study is needed to understand its effects on aggregation mechanism in vivo.

Summary: Protein-protein interactions play a crucial role in biological processes and are attractive drug targets for various human diseases. PCAs offer a direct and versatile approach to study PPIs, enabling the detection of interactions in their native cellular environment. They are valuable tools for large-scale experiments exploring PPI networks and drug screening.
