Bufotalin induces ferroptosis in non-small cell lung cancer cells by facilitating the ubiquitination and degradation of GPX4

Wen Zhang et al.

Summary: In this study, we reported for the first time that Bufotalin (BT), a natural small molecule, is a glutathione peroxidase 4 (GPX4) inhibitor that can trigger ferroptosis in non-small cell lung cancer cells. BT induces ferroptosis and lipid peroxidation by accelerating GPX4 degradation and increasing intracellular Fe2+.

Emerging role of ferroptosis in breast cancer: New dawn for overcoming tumor progression

Shiyao Sui et al.

Summary: Breast cancer is a significant threat to women's health, and ferroptosis may offer a new direction for breast cancer treatment. However, the complete appearance of ferroptosis in breast cancer and its potential combination with traditional anti-breast cancer drugs remain uncertain. This review explores the roles of various factors associated with ferroptosis in breast cancer, provides evidence of ferroptosis induction by traditional drugs and elimination of breast cancer cells by ferroptosis inducers, and discusses the prospects and potential obstacles of using ferroptosis inducers in breast cancer therapy.

Ferroptosis and its Role in Gastric Cancer

Renjun Gu et al.

Summary: Gastric cancer is a common type of cancer with surgery as the main treatment method. However, it is expensive and painful, and patients often experience recurrence. Ferroptosis, a form of cell death dependent on iron, provides a new perspective in cancer therapy. This review summarizes the latest findings on ferroptosis regulation in gastric cancer, discusses the potential of drugs and genes in inducing ferroptosis for treatment and drug resistance, explores the relationship between ferroptosis and the tumor microenvironment in gastric cancer, and suggests areas for future research to accelerate the clinical utility of ferroptosis induction as a treatment strategy for gastric cancer.

Summary: Ferroptosis is an iron-dependent, non-apoptotic form of regulated cell death caused by lipid peroxidation. Lipoxygenase is the main promoter of ferroptosis, while GPX4 is the central repressor of this process.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Broadening horizons: the role of ferroptosis in cancer

Xin Chen et al.

Summary: Ferroptosis is an iron-dependent form of regulated cell death driven by excessive lipid peroxidation, and can be induced by various agents to suppress tumor growth while also potentially triggering inflammation-associated immunosuppression. The extent of ferroptosis's impact on tumor biology and its interactions with other signaling pathways are still under investigation.

Ferroptosis in the tumor microenvironment: perspectives for immunotherapy

Huanji Xu et al.

Summary: Targeting ferroptosis as a new approach for anticancer therapy shows potential as it induces lipid peroxidation in cancer cells. Understanding how immune cells in the tumor microenvironment respond to lethal lipid peroxides may uncover new possibilities for cancer immunotherapy. This study provides an overview of the metabolic processes involved in ferroptosis and explores its roles in cancer immunity, highlighting the potential for targeted therapy.
