One in five patients with Duchenne muscular dystrophy dies from other causes than cardiac or respiratory failure

Summary: This nationwide study in Sweden found that the point prevalence of adult patients with DMD was 3.2 per 100,000 adult males and the birth prevalence was 19.2 per 100,000 male births. The leading cause of death was cardiopulmonary issues, accounting for 79.9% of deaths, while non-cardiopulmonary causes such as injury-related pulmonary embolism, gastrointestinal complications, stroke, and unnatural deaths were also significant factors in mortality among DMD patients.

Summary: Diabetes mellitus is a systemic metabolic disorder associated with mitochondrial dysfunction, and targeting mitochondrial dysfunction with alisporivir, a non-immunosuppressive cyclosporin derivative, can alleviate the development of mitochondrial dysfunction in the diabetic heart. Alisporivir appears to act as a mitochondria-targeted metabolic reprogramming agent, attenuating oxidative damage and stimulating mitophagy in cardiomyocytes of diabetic mice.

The therapy suppresses MPT pore opening, improves calcium homeostasis, but also inhibits mitochondrial biogenesis.
