Mitochondrial disorders of the OXPHOS system

Erika Fernandez-Vizarra et al.

Summary: Mitochondrial disorders are common inborn errors of metabolism, primarily due to dysfunction of the oxidative phosphorylation system (OXPHOS). Around half of diagnosed cases have a known genetic cause, often involving pathogenic variants in genes encoding structural subunits or factors directly involved in ETC assembly.

Structure and Mechanism of Respiratory III-IV Supercomplexes in Bioenergetic Membranes

Peter Brzezinski et al.

Summary: In aerobic organisms, the free energy from electron transfer in the respiratory chain is converted into a proton electrochemical gradient across the membrane for energy storage. Cytochrome c oxidase and cytochrome bc1 combine to form supercomplexes in many organisms, facilitating energy conversion through complex mechanisms and structures.

Protein import in mitochondria biogenesis: guided by targeting signals and sustained by dedicated chaperones

Anna-Roza Dimogkioka et al.

Summary: Mitochondria play a central role in cellular metabolism, with 99% of their proteins encoded by nuclear genes and imported into the organelle through well-studied pathways. Proper folding of these proteins is ensured by a system of chaperones and co-chaperones, critical for maintaining cell homeostasis and response to stress. Understanding the targeting signals and folding mechanisms in mitochondria remains a key area of research.
