Selenium as an emerging versatile player in heterocycles and natural products modification

Summary: The pharmacological activities of organoselenium compounds are related to their ability to scavenge and induce reactive oxygen species (ROS), their oxidative properties, and their Se(0) release property. Incorporating selenium into small molecules, especially heterocycles and natural products, has great potential for altering the potency and selectivity of these molecules. Selenium will thus be crucial in drug discovery in the near future. This article summarizes the effects of different organoselenium species on cellular oxidative stress levels and correlates the structural properties of selenium-containing heterocycles and natural product derivatives with their biological activities and therapeutic applications, providing guidance for rational design of selenium-containing drugs.

Molecular dynamics simulations and in vivo studies in a murine model of acute pancreatitis supported the promising efficacy of these compounds in improving health status and reducing pancreatic damage.

Summary: A series of N,N'-disubstituted selenoureas with an adamantane fragment have been synthesized in yields ranging from 23-75%. Improvements have been made in the isolation and purification methods for aromatic isoselenocyanates. The refined chemical shift of the C=Se carbon nucleus in the C-13 NMR spectra of selenoureas indicates their potential as inhibitors of human soluble epoxide hydrolase, targeting both the epoxide hydrolase (sEH-H) and phosphatase domains (sEH-P).

Activity of Hydroxy Derivatives of Chalcones toward Superoxide Anion Radical

Summary: New hydroxy derivatives of chalcones have been found to exhibit antiradical activity towards superoxide anion radical (O-2(center dot-)) for the first time, with chalcone containing a 2,6-di-tert-butylphenol fragment showing higher activity under adrenaline autooxidation conditions in alkaline medium. Chalcone with two non-shielded OH groups showed the highest trapping activity in enzymatic systems and SOD-protective activity. The inhibiting activity of hydroxy derivatives of chalcones towards O-2(center dot-) is in line with in silico predictions and may decrease the probability of oxidative stress development.
