Mechanism of CFTR correction by type I folding correctors

Karol Fiedorczuk et al.

Summary: Small molecule chaperones have been widely used as therapeutics for diseases caused by protein misfolding. CFTR correctors are the most successful examples, as they can revert folding defects and are used to treat cystic fibrosis. Cryo-electron microscopy structures reveal that these correctors stabilize TMD1 of CFTR, preventing premature degradation and rescuing disease-causing mutations.

Inverse Molecular Docking Study of NS3-Helicase and NS5-RNA Polymerase of Zika Virus as Possible Therapeutic Targets of Ligands Derived from Marcetia taxifolia and Its Implications to Dengue Virus

Cristian Buendia-Atencio et al.

Summary: Dengue and Zika are mosquito-borne diseases that impact public health in tropical and subtropical countries. There are currently no effective therapeutic agents for these pathologies. This study evaluated potential inhibitors from compounds obtained from Marcetia taxifolia through molecular docking, showing that most ligands had better affinity for ZIKV-NS3-helicase than for ZIKV-NS5-RNA polymerase.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

A Review of Trikafta: Triple Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) Modulator Therapy

Anas Zaher et al.

Summary: Cystic fibrosis is a potentially fatal genetic disease, but the FDA-approved drug Trikafta has shown significant efficacy in reducing mortality and morbidity rates in CF patients. By increasing the activity of the CFTR protein, Trikafta outperforms currently available therapies in lung function, quality of life, sweat chloride reduction, and pulmonary exacerbation reduction. Continuing evaluation of patient data is necessary to confirm the long-term safety and efficacy of CFTR modulator therapy.
