Glycogen-based pH and redox sensitive nanoparticles with ginsenoside Rh2 for effective treatment of ulcerative colitis

Yu Xu et al.

Summary: The purpose of this study was to develop pH and redox sensitive nanoparticles for effective treatment of ulcerative colitis (UC) using ginsenoside Rh-2. The study demonstrated that the nanoparticles showed excellent stability and high encapsulation efficiency for Rh-2, and exhibited pH and redox dual response release behavior. In addition, the nanoparticles demonstrated strong anti-inflammatory activity and improved the intestinal flora in mice. Overall, this study provides a promising strategy for the effective treatment of UC.

Phagosomal signalling of the C-type lectin receptor Dectin-1 is terminated by intramembrane proteolysis

Torben Mentrup et al.

Summary: Dectin-1 plays a critical role in recognizing fungal pathogens through pattern recognition receptors on immune cells. Activation of Dectin-1a results in the formation of a stable receptor fragment lacking the ligand binding domain, which contributes to signal transduction in phagosomal membranes and is terminated by intramembrane proteases SPPL2a and 2b.

Nanoparticle Internalization Promotes the Survival of Primary Macrophages

Bader M. Jarai et al.

Summary: This study demonstrates that nanoparticle dosing and cellular internalization can significantly enhance the survival of macrophages. It also shows that poly(ethylene glycol)-based nanoparticle treatment can extend cell viability and increase the expression of prosurvival proteins in macrophages. These findings have important implications for drug screening, vaccine studies, and cell therapies.

Macrophages-targeting mannosylated nanoparticles based on inulin for the treatment of inflammatory bowel disease (IBD)

Qijuan Sun et al.

Summary: A novel nanocomposite was developed in this study for real-time imaging and therapeutic targeting of activated macrophages in the colon. The nanocomposite, formed by the covalent conjugation of Man-NPs with CDs, showed high drug casing density, negative charge, and specific uptake by inflamed macrophages, suggesting potential for treating inflammatory bowel disease.

Nanoparticle size distribution quantification from transmission electron microscopy (TEM) of ruthenium tetroxide stained polymeric nanoparticles

Brian K. Wilson et al.

Summary: The hypothesis states that DLS-generated PSD of polymer-stabilized nanoparticles is dependent on the optimization parameters used for inversion solution fit. By comparing TEM images with DLS results, the accuracy of the DLS PSD and polydispersity can be determined. The experiments show that DLS consistently overestimates the standard deviation of the PSD compared to TEM measurements.

Uptake and Intracellular Trafficking Studies of Multiple Dye-Doped Core-Shell Silica Nanoparticles in Lymphoid and Myeloid Cells

Federica Sola et al.

Summary: The study on NT(B)700 NPs showed efficient uptake process in cells with the involvement of mitochondria and lysosome pathways, without inducing significant cytotoxic effects. The NPs did not show a preferred internalization route, and despite localizing mainly in mitochondria, they did not significantly increase intracellular ROS levels, which are known inducers of apoptosis. Lymphoid and myeloid cells were able to release the NPs, indicating their biosafety.
