Urinary C5b-9 as a Prognostic Marker in IgA Nephropathy

Byung Chul Yu et al.

Summary: In this study, the potential of urinary C5b-9 as a prognostic biomarker for IgAN was evaluated. The results showed a positive correlation between baseline urinary C5b-9 levels and proteinuria, as well as a correlation between changes in urinary C5b-9 levels and changes in proteinuria and eGFR after treatment. However, baseline urinary C5b-9 levels were not a significant independent factor for predicting treatment response.

Results from a nationwide retrospective cohort measure the impact of C3 and soluble C5b-9 levels on kidney outcomes in C3 glomerulopathy

Sophie Chauvet et al.

Summary: C3 glomerulopathy (C3G) is a rare complement-mediated disease. The prognosis of this disease is associated with age, kidney function and genetic variants in complement genes. Blood biomarker analysis can predict the kidney outcome of patients.

Factor H related proteins modulate complement activation on kidney cells

Brandon Renner et al.

Summary: This study demonstrates that the Factor H-related proteins have unique effects on different cell types within the kidney, with FHR E causing glomerular complement dysregulation in vivo. This has important implications for understanding the pathogenesis of glomerular diseases.

Clinical significance of glomerular C3 deposition in primary membranous nephropathy

Ozgur Akin Oto et al.

Summary: This study found that the extent of glomerular C3 deposition is associated with the development of kidney failure in patients with primary membranous nephropathy. Patients with high intensity C3 deposition had lower eGFR, higher proteinuria, and were more likely to experience kidney failure compared to those with low intensity C3 deposition.

Inherited Kidney Complement Diseases

Mathieu Lemaire et al.

Summary: In the past 20 years, there have been significant advances in diagnosing and treating genetic kidney diseases caused by complement dysregulation. One major area of progress has been in understanding the genetic basis and pathophysiology of aHUS and C3-dominant glomerulopathies, leading to the development of innovative therapies that have greatly improved patient outcomes. Key genes associated with these diseases have been identified, and future directions for research are being explored.

Functional characterization of 105 factor H variants associated with aHUS: lessons for variant classification

Hector Martin Merinero et al.

Summary: Atypical hemolytic uremic syndrome (aHUS) is a life-threatening thrombotic microangiopathy mainly caused by pathogenic variants in genes encoding complement components and regulators. Accurate classification of FH gene CFH variants remains challenging due to lack of functional studies, emphasizing the importance of functional assays in interpreting FH variants.

Expanding Horizons in Complement Analysis and Quality Control

Ashley Frazer-Abel et al.

Summary: Complement not only plays a crucial role in host microbial defense but also modulates adaptive immune responses by modifying T and B cell reactivity. Dysregulation and overactivation of the complement system in clinical practice are major causes of various inflammatory and autoimmune diseases. Efforts to standardize complement testing have improved consistency across laboratories and provided a comprehensive insight into deficiencies and activation state of the complement system for better defining disease severity, evolution, and response to therapy.

Complement Activation Fragments Are Increased in Critically Ill Pediatric Patients with Severe AKI

Erin K. Stenson et al.

Summary: This study investigated the association between complement activation fragments and severity of AKI in critically ill children. It found that levels of various complement fragments increase with the severity of AKI, and identified specific fragments like plasma C4a as potential risk factors for adverse outcomes. Further research is needed to explore the role of complement activation in critically ill children at risk of AKI.
