Elp3-mediated codon-dependent translation promotes mTORC2 activation and regulates macrophage polarization

Dawei Chen et al.

Summary: This study reveals the involvement of translational reprogramming in macrophage polarization, with a focus on the role of the Elongator complex subunit Elp3. Elp3 expression is regulated by different activating signals, with M1 signals downregulating its expression and M2 signals upregulating it. The metabolic reprogramming associated with M2 macrophage polarization is dependent on Elp3 and the translation of specific genes. Additionally, Elp3 plays a role in mTORC2 activation and the initiation of Wnt-driven tumor in the intestine by maintaining a pool of M2-like tumor-associated macrophages.

CD40-mediated immune cell activation enhances response to anti-PD-1 in murine intrahepatic cholangiocarcinoma

Laurence P. Diggs et al.

Summary: Stimulating antigen-presenting cells such as macrophages and dendritic cells through CD40 agonist significantly enhances the response to anti-PD-1 therapy in intrahepatic cholangiocarcinoma (iCCA) models, leading to a greater reduction in tumor burden. The combination of CD40 agonist and PD-1 inhibitor, along with chemotherapy, shows promising results in improving therapeutic efficacy and overall survival.

Gut Microbiome Directs Hepatocytes to Recruit MDSCs and Promote Cholangiocarcinoma

Qianfei Zhang et al.

Summary: The study demonstrates that gut dysbiosis can lead to the accumulation of PMN-MDSCs, promoting liver cancer development. However, treatment with neomycin can block this process and inhibit tumor growth, even in the absence of liver disease or colitis.

Myeloid-Derived Suppressor Cells: A Propitious Road to Clinic

Amit Grover et al.

Summary: MDSCs are important regulators of immune responses in cancer, and their association with negative clinical outcomes makes them attractive therapeutic targets. However, the complex nature of MDSC biology presents a substantial challenge for therapeutic targeting. This review discusses the challenges and potential solutions in targeting MDSCs for therapeutic intervention.

MDSC: Markers, development, states, and unaddressed complexity

Samarth Hegde et al.

Summary: MDSCs are a widely discussed biological entity in immunology, commonly describing cells that arise during chronic inflammation with T cell immunosuppressive functions. Currently lacking clear definitions and a unifying framework across pathologies, we propose a framework based on activation signals to classify MDSCs as discrete cell states. Developing this knowledge of myeloid states across pathological conditions may transform how diseases are grouped and treated.

METTL1-m7G-EGFR/EFEMP1 axis promotes the bladder cancer development

Xiaoling Ying et al.

Summary: The study reveals that METTL1 is highly expressed in bladder cancer (BC) and correlates with poor patient prognosis. Silencing METTL1 inhibits proliferation, migration, and invasion of BC cells by altering the expression of certain target genes, such as EGFR/EFEMP1, through m(7)G tRNA modification. Forced expression of EGFR/EFEMP1 partially rescues the effects of METTL1 deletion on BC cells, highlighting the potential therapeutic targets in BC treatment.

METTL1 promotes hepatocarcinogenesis via m7G tRNA modification-dependent translation control

Zhihang Chen et al.

Summary: The N-7-methylguanosine (m(7)G) modification is a common transfer RNA (tRNA) modification in humans. Elevated levels of METTL1 and WDR4 in hepatocellular carcinoma (HCC) are associated with advanced tumor stages and poor patient survival. Mechanistically, METTL1-mediated tRNA m(7)G modification promotes the translation of target mRNAs with higher frequencies of m(7)G-related codons, providing new insights into the role of misregulated tRNA modifications in liver cancer.

N7-Methylguanosine tRNA modification enhances oncogenic mRNA translation and promotes intrahepatic cholangiocarcinoma progression

Zihao Dai et al.

Summary: Intrahepatic cholangiocarcinoma (ICC) shows upregulation of N-7-methylguanosine (m(7)G) tRNA modification and its methyltransferase complex components, METTL1 and WDR4, which are associated with poor prognosis. METTL/WDR4 play a critical role in promoting ICC cell survival and progression by selectively regulating the translation of oncogenic transcripts through m(7)G-tRNA-decoded codon-frequency-dependent mechanisms. Studies with overexpression and knockout mouse models demonstrate the crucial oncogenic function of Mettl1-mediated m(7)G tRNA modification in promoting ICC tumorigenesis and progression in vivo.

METTL1/WDR4-mediated m7G tRNA modifications and m7G codon usage promote mRNA translation and lung cancer progression

Jieyi Ma et al.

Summary: This study found that the expression levels of tRNA N7-methylguanosine (m7G) methyltransferase complex components METTL1 and WDR4 are significantly elevated in human lung cancer samples and negatively associated with patient prognosis. METTL1 promotes lung cancer growth and invasion through regulation of m7G tRNA modifications, and tRNA modifications and codon usage play an essential function in mRNA translation regulation.

Myeloid-derived suppressor cells in the era of increasing myeloid cell diversity

Filippo Veglia et al.

Summary: MDSCs are pathologically activated cells with potent immunosuppressive activity, closely associated with poor clinical outcomes in diseases like cancer. Recent studies have identified key distinctions between MDSCs and classical neutrophils and monocytes, providing new insights into therapeutic targeting for cancer and autoimmune diseases through understanding their genomic and metabolic characteristics. Emerging data also suggest potential involvement of MDSCs in pregnancy, neonatal biology, and COVID-19.
