It revealed that peptide presentation in lung adenocarcinoma is not predictable by mRNA expression or translation efficiency and may be driven by post-translational mechanisms. Vaccination with peptides presented by lung adenocarcinoma induced CD8(+) T cell responses.

Studies have shown that a significant portion of PDAC patients have predicted high-affinity MHC class I-restricted neoepitopes, and modulation of the CD155/TIGIT axis can promote immune evasion in PDAC. In preclinical models, a combination immunotherapy targeting TIGIT/PD-1 plus CD40 agonism has shown promising anti-tumor responses.

However, the subdominant T cell response does not preferentially benefit from ICB due to a dysfunctional subset of TCF1(+) cells marked by CCR6 and Tc17 differentiation.

The decrease in conventional type I dendritic cells (cDC1) in dLN contributes to failed anti-tumor immunity by reducing the reservoir of TCF-1(+) CD8(+) T cells.
