Development and inter-laboratory validation of the VISAGE enhanced tool for age estimation from semen using quantitative DNA methylation analysis

Summary: The analysis of DNA methylation is a well-established method for age estimation, with various assays available for somatic tissues like blood. However, age estimation from semen requires different DNA methylation markers. A new assay using massively parallel sequencing to analyse 13 candidate CpG sites in two multiplex PCRs has been developed and validated for age estimation from semen in forensic investigations, showing robust results.

Extended kinship analysis of historical remains using SNP capture

Summary: DNA-assisted identification of historical remains requires the genetic analysis of highly degraded DNA and a comparison to DNA from known relatives. In this study, SNP capture combined with next-generation sequencing on a benchtop platform was used to target single nucleotide polymorphisms (SNPs) and estimate distant kinship. The results showed that low-coverage SNP data can be recovered from degraded skeletal samples, enabling accurate kinship predictions.
