TRPC6 inhibits renal tubular epithelial cell pyroptosis through regulating zinc influx and alleviates renal ischemia-reperfusion injury

Skin damage induced by zinc oxide nanoparticles combined with UVB is mediated by activating cell pyroptosis via the NLRP3 inflammasome-autophagy-exosomal pathway

Summary: Our study found that co-exposure to UVB and zinc oxide nanoparticles (ZnONPs) leads to activation of the NLRP3 inflammasome and pyroptosis in keratinocytes. Additionally, the exposure to both UVB and ZnONPs disrupts cellular autophagy and increases cell exosome release. In vivo, UVB and ZnONPs exposure causes skin toxicity, as indicated by histological injury, increased skin thickness, and transepidermal water loss. Notably, the NLRP3 inflammasome-mediated pyroptosis is also activated during exposure. Topical application of pterostilbene (PT) attenuates NLRP3 inflammasome activation and pyroptosis by reducing ROS generation and mitochondrial ROS (mtROS) levels.

Association of zinc administration with growth suppression of intracranial aneurysms via induction of A20

Summary: This study demonstrated that zinc administration can inhibit the growth of intracranial aneurysms in rats by preserving the internal elastic lumen, reducing macrophage infiltration, and increasing the expression of the antiinflammatory signaling protein A20. Zinc treatment may prevent the growth of rat IAs by inducing A20-attributed inactivation of the NF-kappa B signaling pathway.

Selective TRPC6 blocker BI-749327 shows potential as a novel and effective drug for treating PAH and PH due to respiratory diseases or hypoxemia, reversing PH by 50% and inhibiting phosphorylation of AKT and mTOR in PASMCs.
