Exerkines in health, resilience and disease

Lisa S. Chow et al.

Summary: This Review summarizes the importance and current state of exerkine research, highlighting the potential roles of exerkines in improving cardiovascular, metabolic, immune, and neurological health. Exerkines are signaling molecules released during exercise, which have the potential for the treatment of cardiovascular disease, type 2 diabetes mellitus, obesity, and to facilitate healthy aging.

Roles of mitochondrial DNA in dynamics of the immune response to COVID-19

Ata Mahmoodpoor et al.

Summary: Mitochondrial DNA plays a crucial role in the immune response to COVID-19, guiding the immune response against pathogens and serving as a valuable indicator for the prognosis and severity of infectious diseases.

Mitochondrial DNA copy number, metabolic syndrome, and insulin sensitivity: Insights from the Sugar, Hypertension, and Physical Exercise studies

Stephanie Y. Yang et al.

Summary: The study evaluated the effect of 6-months of exercise intervention on mtDNA-CN measured in blood. Although no significant relationship was found between exercise intervention and mtDNA-CN change, there was a significant association between mtDNA-CN and metabolic syndrome, as well as a significant association between higher mtDNA-CN and higher insulin sensitivity.

Common mitochondrial haplogroups as modifiers of the onset-age for critical COVID-19

Daniel Vazquez-Coto et al.

Summary: Mitochondrial variants, particularly haplogroup H, may serve as predictors of COVID-19 severity, with haplogroup H having a protective effect and the 16223T variant increasing the risk of early-onset critical COVID-19.

Common human genetic variants of APOE impact murine COVID-19 mortality

Benjamin N. Ostendorf et al.

Summary: The common variants of the APOE gene may contribute to the heterogeneous outcomes of COVID-19. Mice with APOE2 or APOE4 variants exhibited rapid disease progression and poor survival outcomes, possibly due to increased viral loads and suppressed antiviral immune responses. The APOE genotype was also found to be associated with survival in patients infected with SARS-CoV-2, suggesting its potential as a predictive factor for adverse outcomes.

Deciphering the genetic and epidemiological landscape of mitochondrial DNA abundance

Sara Hagg et al.

Summary: This study investigated the factors influencing mitochondrial DNA (mtDNA) abundance, revealing that it is higher in women compared to men and negatively correlated with age, smoking exposure, BMI, frailty index, and certain blood biochemistry markers. Genome-wide association study identified 50 regions significantly associated with mtDNA abundance, indicating the involvement of genes related to immunity, cancer, and mitochondrial function. The SNP-heritability of mtDNA abundance was estimated to be around 8%, and mtDNA abundance was found to be associated with leukemia, hematologic diseases, and hypertension.

Genetic mechanisms of critical illness in COVID-19

Erola Pairo-Castineira et al.

Summary: In critically ill patients with COVID-19, host genetic variants associated with antiviral defense mechanisms and inflammatory organ damage were identified through a genome-wide association study. These genetic signals provide potential targets for therapeutic development and repurposing of existing drugs. Large-scale randomized clinical trials will be essential to confirm the effectiveness of targeted treatment strategies.

Mitochondrial phenotypes in purified human immune cell subtypes and cell mixtures

Shannon Rausser et al.

Summary: Using a high-throughput mitochondrial phenotyping platform, this study quantified multiple mitochondrial features among different immune cell subtypes in humans, revealing natural variations in mtDNA copy number, citrate synthase, and respiratory chain enzymatic activities in various cell types. The results showed that cell type distributions and contaminating platelets can confound mitochondrial measures. Age- and sex-related differences in mitochondrial features were observed, with age-related elevation in mtDNAcn often masked in mixed PBMCs. A proof-of-concept study in a single individual also demonstrated cell type differences and week-to-week changes in mitochondrial activities. Validation and mechanistic studies are needed to further explore these findings.

Blood mitochondrial DNA copy number: What are we counting?

Martin Picard

Summary: The mitochondrial DNA copy number (mtDNAcn) is a measure of mitochondrial health, but its interpretation may not be consistent. Blood-derived mtDNAcn is influenced by the number of leukocytes and platelets, and may be affected by variations in blood cell types.
