Label-Free Quantitative Proteomics Analysis of the Sorafenib Resistance in HepG2 Cells

Summary: In this study, a label-free quantitative proteomics method was used to investigate differentially expressed proteins between sorafenib-resistant and sorafenib-sensitive hepatocellular carcinoma cells. The results suggest that dysregulated metabolic processes may play a significant role in drug resistance. Further experiments confirmed that MGST1 is closely associated with sorafenib resistance, and knockdown of MGST1 increased sensitivity to sorafenib treatment in resistant cells, indicating its potential as a therapeutic target for overcoming sorafenib resistance in HCC.

Vinculin orchestrates prostate cancer progression by regulating tumor cell invasion, migration, and proliferation

Summary: This study evaluated the potential role of vinculin (VCL) in prostate cancer (PCa) progression through in vitro and in vivo experiments. The methylation level of the VCL promoter in PCa was found to be downregulated with age and nodal metastasis progression. Knockdown of VCL significantly changed cell morphology, inhibited migration, invasion, movement, and colony formation, as well as suppressed tumor growth in vivo. These findings suggest that VCL could be a potential target for PCa prognosis and treatment.

Derivation and Validation of a Prognostic Model for Cancer Dependency Genes Based on CRISPR-Cas9 in Gastric Adenocarcinoma

Summary: This study utilized CRISPR-Cas9 tool to investigate the expression and prognostic significance of Cancer Dependency Map genes in gastric adenocarcinoma, identifying 147 upregulated and 99 downregulated differentially expressed CDMs. A prognostic model based on ten CDMs was constructed, showing good predictive performance. Additionally, PWP2 gene was found to promote cell invasion and migration in gastric adenocarcinoma through the EMT signaling pathway.

Summary: Loss of function of tumor suppressor proteins in cancer cells is often the result of genetic mutations. Protein Phosphatase 2A (PP2A) is a tumor suppressor that is inactivated by endogenous inhibitors, leading to sustained activation of mitogenic signaling pathways. One of these inhibitors, SET, interacts with PP2A and is associated with high expression in various cancers, suggesting disruption of SET-PP2A interaction as a potential therapeutic strategy.
