Cancer Chemotherapy-Induced Sinus Bradycardia: A Narrative Review of a Forgotten Adverse Effect of Cardiotoxicity

Juan Tamargo et al.

Summary: Cardiotoxicity is a common adverse effect of anticancer drugs, including targeted drugs, and can increase morbidity and mortality in cancer patients. While most attention has been given to heart failure, myocardial ischemia, hypertension, thromboembolism, QT prolongation, and tachyarrhythmias caused by these drugs, the proarrhythmic effect of sinus bradycardia remains underestimated. Sinus bradycardia is of interest due to its association with QT prolongation and its increasing incidence with aging population and age-related diseases like cancer.

Lapatinib induces mitochondrial dysfunction to enhance oxidative stress and ferroptosis in doxorubicin-induced cardiomyocytes via inhibition of PI3K/AKT signaling pathway

Lei Sun et al.

Summary: This study aimed to explore the mechanism involved in the synergistic effect of lapatinib (LAP) in doxorubicin (DOX)-induced cardiotoxicity. The results showed that LAP aggravated cell injury, apoptosis, and mitochondrial dysfunction induced by DOX. Additionally, LAP in combination with DOX induced oxidative stress and ferroptosis. The activation of PI3K/AKT signaling reversed the detrimental effects of LAP on DOX-induced cardiotoxicity.

Myocardial injury detected by T1 and T2 mapping on CMR predicts subsequent cancer therapy-related cardiac dysfunction in patients with breast cancer treated by epirubicin-based chemotherapy or left-sided RT

Enver Tahir et al.

Summary: This study analyzed the serial laboratory and cardiac magnetic resonance imaging (CMR) parameters to predict cancer therapy-related cardiac dysfunction (CTRCD) after epirubicin-based chemotherapy compared with left-sided radiotherapy. The results showed that T1 and T2 relaxation times increased after epirubicin-based chemotherapy, but not after left-sided radiotherapy. Combined use of increased T1 and reduced left ventricular ejection fraction (LVEF) had good sensitivity and specificity in predicting CTRCD.

Ferroptosis triggered by dihydroartemisinin facilitates chlorin e6 induced photodynamic therapy against lung cancer through inhibiting GPX4 and enhancing ROS

Ning Han et al.

Summary: Photodynamic therapy (PDT) is a noninvasive, low toxicity, and photo-selective treatment method, but malignant cells may resist it. This study found that chlorin e6 (Ce6) mediated PDT showed drug resistance in lung cancer cells (LLC), which may be associated with PDT-induced DNA damage response (DDR). Dihydroartemisinin (DHA) was found to down-regulate glutathione peroxidase 4 (GPX4) and enhance the efficacy of PDT. This study provides a theoretical basis for regulating the intratumoral redox system and improving PDT efficacy against lung cancer.

New progress in elucidating the relationship between cancer therapy and cardiovascular toxicity

Chen Wu et al.

Summary: Onco-cardiology is an emerging field that connects cancer with cardiovascular injury, aiming to understand the mechanisms of cardiac damage to improve the quality of life for cancer survivors. Current cancer therapies have been found to have latent cardiovascular toxicity, leading to adverse cardiovascular events such as heart failure, hypertension, coronary heart diseases, arrhythmias, and thromboembolism, even years after treatment has concluded. Therefore, there is a growing need to focus on revealing the mechanism of cancer treatment-related cardiovascular toxicity to guide prevention, diagnosis, and treatment of cardiovascular diseases in cancer survivors.

Anna Yu. Korshunova et al.

Summary: Cardiovascular disease is the leading cause of death in developed countries, with coronary heart disease being the main culprit. Myocardial reperfusion is crucial for preventing cell death after ischemia, but it can also cause further damage. Understanding the mechanisms of regulated cell death, with a focus on anti-apoptotic proteins like BCL2, is essential for modern cardioprotective therapies in treating myocardial ischemia-reperfusion injury.
