ABCB1 as a potential beneficial target of midostaurin in acute myeloid leukemia

Summary: This study evaluated the clinical relevance of ABCB1 in AML patients and found that overexpression of ABCB1 was associated with adverse prognosis. Midostaurin, as an ABCB1 inhibitor, increased the accumulation of anthracyclines in AML patients and promoted apoptosis in leukemic cells. Additionally, miR-9 was identified as a useful prognostic marker in AML.

Additionally, WS-898 showed promising results in in vivo PTX sensitization without causing obvious toxicity.

In vitro experiments showed that the nanoparticles were able to down-regulate P-glycoprotein expression and improve anticancer efficacy. Additionally, NIR laser irradiation induced nanoparticles to generate reactive oxygen species, effectively inhibiting multidrug resistance and metastasis in breast cancer.

An insight into the structure of 5-spiro aromatic derivatives of imidazolidine-2,4-dione, a new group of very potent inhibitors of tumor multidrug resistance in T-lymphoma cells

Summary: A series of newly synthesized compounds were explored for their potential as potent inhibitors of cancer multidrug resistance efflux pumps. Particularly, the spirofluorene derivatives with amine alkyl substituents at position 1 showed the strongest P-glycoprotein inhibitory activity in MDR T-lymphoma cells, with compounds 7 and 12 being 100-fold more potent than verapamil. Crystallography-supported pharmacophore-based SAR analysis suggested specific structural properties contributing to this excellent cancer MDR-inhibitory action.
