1-Nitropyrene disrupts testosterone biogenesis via AKAP1 degradation promoted mitochondrial fission in mouse Leydig cell

Wei-wei Zhang et al.

Summary: The study found that 1-NP disrupts steroidogenesis in MLTC-1 cells by promoting ROS-mediated AKAP1 degradation and excessive mitochondrial fission. It also showed that cellular ROS and oxidative stress were elevated in 1-NP-exposed cells. The antioxidant NAC alleviated the effects of 1-NP by reducing ROS levels and preventing AKAP1 degradation. This study provides important insights into the endocrine disruption caused by 1-NP and offers a theoretical basis for public health prevention.

Summary: The study found that anoxia-reoxygenation and cadmium have different effects on H2O2 emission in fish liver mitochondria, depending on the substrate used, leading to different response patterns. Additionally, anoxia-reoxygenation inhibits mitochondrial respiration in a substrate-dependent manner.

Ribosome profiling reveals ribosome stalling on tryptophan codons and ribosome queuing upon oxidative stress in fission yeast

Angela Rubio et al.

Summary: Translational control is crucial in response to stress, with different stresses inducing common and specific translational responses in cells. The transcription factor Fil1 plays a key role in the cellular response to most stresses, while many mRNAs encoding proteins for ribosome biogenesis are translationally downregulated. Additionally, tryptophan codon stalling upon oxidative stress affects translation elongation and contributes to the ISR-mediated inhibition of initiation.

Ribosome states signal RNA quality control

Karole N. D'Orazio et al.

Summary: Eukaryotic cells integrate multiple quality control responses during protein synthesis, signaled by slow or stalled elongating ribosomes. Depending on the nature of the delay, the signal may lead to translational repression, messenger RNA decay, ribosome rescue, and/or nascent protein degradation. The structure and composition of an elongating ribosome in a troubled state determine the downstream quality control pathways involved, highlighting the intersecting pathways between RNA decay and ribosome rescue.

Characteristics and unique sources of polycyclic aromatic hydrocarbons and nitro-polycyclic aromatic hydrocarbons in PM2.5 at a highland background site in northwestern China

Lulu Zhang et al.

Summary: PAHs and NPAHs were observed in PM2.5 at a background site in the northwestern highlands of China, reaching peak concentrations in winter. Vehicle emissions were identified as the main source, with contributions from combustion sources. The burning of Tibetan barley straw was found to be a seasonal contributor to air pollution in northwestern China, participating in the atmospheric transport of pollutants driven by the East Asian monsoon.

Global phosphoproteomics pinpoints uncharted Gcn2-mediated mechanisms of translational control

Ladislav Dokladal et al.

Summary: Gcn2 protein kinase mediates cellular adaptations to amino acid limitation by phosphorylating eIF2 alpha, as well as targeting auxiliary effectors such as eIF2 G protein complex for translational control. This mechanism contributes to the inhibition of translation initiation in amino-acid-starved cells and plays a more intricate role in translation control than previously understood.

Structure of Gcn1 bound to stalled and colliding 80S ribosomes

Agnieszka A. Pochopien et al.

Summary: The Gcn pathway is highly conserved in eukaryotes, with Gcn1 serving as an essential effector protein for Gcn2 kinase to regulate global translation in response to stress. The structure of Gcn1 interacting with stalled and colliding ribosomes was revealed through cryo-electron microscopy, shedding light on the activation of Gcn2. This study provides important insights into the molecular mechanism of Gcn pathway activation and its implications in ribosome-associated quality control pathways.

SLFN2 protection of tRNAs from stress-induced cleavage is essential for T cell-mediated immunity

Tao Yue et al.

Summary: ROS in activated T cells triggers an oxidative stress response leading to translation repression, which is countered by SLFN2 binding tRNAs. SLFN2 deficiency results in tRNA fragment accumulation, inhibiting translation and promoting stress-granule formation.

Hua-Long Zhu et al.

Summary: Gestational exposure to environmental stress can lead to fetal growth restriction (FGR) with reduced levels of melatonin (MT) in the placenta. MT offers protective effects against FGR by inhibiting BNIP3-dependent mitophagy, potentially serving as a therapeutic agent against environmental stress-induced developmental toxicity.
