Caspase-1 and Gasdermin D Afford the Optimal Targets with Distinct Switching Strategies in NLRP1b Inflammasome-Induced Cell Death

Xiang Li et al.

Summary: This study investigates the regulatory mechanism of inflammasome signaling and its correlation with COVID-19. The expression levels of caspase-1 and GSDMD are found to individually switch cell death modes, with caspase-1 and GSDMD presenting different thresholds and pathway choices. Additionally, caspase-1 and GSDMD can ignite the cooccurrence of pyroptosis and apoptosis. A seesaw model is proposed to describe the death states controlled by caspase-1 or GSDMD. This study sheds new light on NLRP1b inflammasome signaling and provides potential clues for disease control strategies.

RIP1-dependent linear and nonlinear recruitments of caspase-8 and RIP3 respectively to necrosome specify distinct cell death outcomes

Xiang Li et al.

Summary: This study reveals the significant impact of RIP1 level on the crosstalk between apoptosis and necroptosis pathways, showing that RIP1 above approximately 1000 mpc leads to necroptosis, while levels above approximately 46,000 mpc inhibit apoptosis and lead to necroptosis alone. The findings provide insights into the diverse roles of RIP1 in governing cell fate decisions.

Dynamical analysis of the fission yeast cell cycle via Markov chain

Sajad Shafiekhani et al.

Summary: The cell cycle is a complex network that can experience fluctuations in protein concentration due to intrinsic noise in gene expression. Understanding how the cell cycle network maintains robust transitions in the presence of these fluctuations is important. By developing a Markov model for the fission yeast cell cycle system, the study quantified the effects of noise on gene and protein activity, predicting the most robust and probable pathway in the cell cycle network. The sensitivity analysis conducted also provided insights on how network perturbations affect transition probabilities between cell cycle phases and cell fates.

Crosstalk between Plk1, p53, cell cycle, and G2/M DNA damage checkpoint regulation in cancer: computational modeling and analysis

Yongwoon Jung et al.

Summary: Different cancer cell lines have varying responses to perturbations, with cells carrying DNA damage checkpoint-related mutations being more sensitive to gene perturbations. A computational model was developed to study the interactions between Plk1, p53, and G2/M DNA damage checkpoint, successfully explaining the phenotypes of different cancer cell lines. The model can be used to predict the effect of potential treatments targeting key regulators on cell cycle progression.

Modeling and analysis of the macronutrient signaling network in budding yeast

Amogh P. Jalihal et al.

Summary: Adaptive modulation of cellular growth state is crucial for survival in fluctuating nutrient environments. The complex nutrient signaling network in eukaryotes, like budding yeast, must have inbuilt robustness to reliably respond to varying conditions. A computational model of this network was developed to reconcile experimental data and predict nutrient responsive transcription factor activities in mutant strains.

Summary: This study proposes a minimal necroptotic circuit based on the CNS pathway and demonstrates the essential structure and parameter characteristics required for oscillation behavior in the circuit. It also classifies different types of oscillation behaviors and explores their robustness, highlighting the importance of understanding oscillations in regulating cell death signaling in the necroptotic network.
