Transcription Factor TWIST1 Integrates Dendritic Remodeling and Chronic Stress to Promote Depressive-like Behaviors

Summary: The study found that the expression of TWIST1 in the brain is positively correlated with depressive behaviors, chronic stress can elevate TWIST1 expression in the medial prefrontal cortex of mice which can be reversed by fluoxetine treatment. Overexpression of TWIST1 increases susceptibility to stress, while knockdown of TWIST1 prevents defective dendrite morphogenesis in mice and alleviates depressive-like behaviors. The prodepressant property of TWIST1 is mediated by the repression of miR-214-PPAR-delta signaling and mitochondrial function, which can be mimicked by genetic and pharmacological inhibition of PPAR-delta.
