Development of a Fast Raman-Assisted Antibiotic Susceptibility Test (FRAST) for the Antibiotic Resistance Analysis of Clinical Urine and Blood Samples

Summary: Human health is facing great risks due to the spread of antimicrobial resistance, which has led to the development of a fast Raman-assisted antibiotic susceptibility test (FRAST). This new method showed an overall agreement of 88.0% with conventional AST, and can further reduce turnaround time by using a gram-staining classification based on linear discriminant analysis of SCRS. By applying FRAST to real clinical analysis, it provides fast and reliable susceptibility tests which can speed up microbiological analysis for clinical practice and facilitate antibiotic stewardship.

Raman microscopy-based quantification of the physical properties of intracellular lipids

Summary: A novel Raman microscopy-based approach was developed for quantifying the physical properties of intracellular lipids under deuterium-labeled fatty acid treatment conditions. The study found that the gauche/trans conformational ratio of labeled fatty acids can be used as an indicator of viscosity and the physical properties of intracellular lipids are robustly preserved due to the buffering role of lipid droplets against excess ratios. This novel approach allows for the observation of organelle lipids' physical properties and quantitative assessment of the subcellular lipid environment.
