Additionally, Pex11 has a Pex19-independent peroxisome-targeting signal (mPTS) that anchors it to the outer leaflet of the peroxisomal membrane. Pex19 acts as a chaperone that protects Pex11 from spontaneous oligomerization and subsequent degradation.

Summary: In the yeast Hansenula polymorpha, the proteins Pex11 and Pex23 family are important for the formation of peroxisome-ER contact sites, which are involved in lipid transfer and peroxisomal membrane expansion. The contact site protein Vps13 was identified as essential for peroxisome biogenesis in cells lacking Pex11, Pex23, or Pex24. Additionally, Vps13 plays a redundant role in lipid transfer from the ER to peroxisomes in cells with reduced peroxisome-ER contact sites.

Yeast as a Model to Find New Drugs and Drug Targets for VPS13-Dependent Neurodegenerative Diseases

Summary: Mutations in VPS13A-D genes in humans lead to rare neurological diseases, such as chorea-acanthocytosis, with unknown pathogenesis and no effective treatment. Yeast is used as a model organism to study the effects of these pathogenic mutations. In recent studies, yeast mutants vps13 Delta and vps13-I2749, serving as models for chorea-acanthocytosis, were used to screen for multicopy and chemical suppressors. Several suppressors were identified, including genes involved in calcium signaling and iron and copper homeostasis. These findings provide insights for further investigation in higher eukaryotes and potential therapeutic targets for VPS13-related diseases.
