Summary: In this study, bio-asymmetric synthesis of D(-)-pantolactone (DPL) from ketopantoyl lactone (KPL) by utilizing a conjugated polyketone reductase from Candida glabrata (Cg/CPR) was achieved with high expression in Escherichia coli. The purified Cg/CPR showed optimal temperature and pH for catalytic activity, with Michaelis-Menten constants Km and Vmax determined for KPL. To prevent spontaneous degradation of KPL, a strategy combining fed-batch addition of whole cell catalyst with substrate semi-continuous feeding was employed, resulting in a high isolated yield of DPL and excellent enantiomeric excess (>99%). The space-time yield of DPL reached 228 g L-1 d-1 after a 13-hour reaction.

This study indicates the promising industrial application prospects of D-Lacs expressed by P. pastoris.

Summary: The ligninolytic strain of white-rot fungus B. adusta CCBAS 930 and its mutants were effective in removing molasses by decolorizing and decomposing melanoidins. Mutant 930-5 showed the highest efficiency in decolorization and decomposition of melanoidin, with a significant increase in HRP-like peroxidase activity in immobilized cultures.

Strengthening the (R)-pantoate pathway to produce D-pantothenic acid based on systematic metabolic analysis

Summary: Through systematic metabolic engineering, higher yielding D-pantothenic acid producing strains were obtained based on previously developed strains. Overexpression of lpd gene, disruption of non-PTS pathway, and enhancement of PTS pathway were conducted to increase precursor accumulation. Blocking pyruvate bypass pathway, enhancing (R)-pantothenic acid pathway, introducing heterologous proteins, and blocking other synthesis pathways significantly increased the yield of D-pantothenic acid to 6.33 g/L.
