Summary: PSMA, which is overexpressed in prostate cancer cells, is a suitable target for specific delivery of antitumor drugs and diagnostic agents. The novel PSMA ligands designed exhibit strong inhibition on PSMA activity, with conjugates showing high affinity to PSMA-expressing tumor cells in vitro. In vivo biodistribution of PSMA-SulfoCy5 and PSMA-SulfoCy7 conjugates with a novel PSMA ligand in xenografts demonstrates good visualization of PSMA-expressing tumors, with no explicit toxicity observed up to 87.9 mg/kg.
