ANGPTL1 attenuates cancer migration, invasion, and stemness through regulating FOXO3a-mediated SOX2 expression in colorectal cancer

Ting-Yu Chang et al.

Summary: This study identified that ANGPTL1 is down-regulated in colorectal cancer (CRC) and is inversely correlated with metastasis and poor clinical outcomes. ANGPTL1 suppresses migration/invasion abilities, the expression of cancer stem cell markers, and sphere formation in CRC cells by enhancing FOXO3a expression. Overexpression of ANGPTL1 reduces liver metastasis, tumor growth, and tumorigenicity in tumor-bearing mice.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Molecular Mechanisms of Colon Cancer Progression and Metastasis: Recent Insights and Advancements

Ahmed Malki et al.

Summary: Colorectal cancer is the second leading cause of cancer-related mortality rates worldwide, with its prevalence still on the rise. Dysregulation of several cellular signaling pathways in CRC leads to malignant phenotypes. Studying signaling pathways helps elucidate the mechanisms of CRC progression and pharmacotherapy.

Exosomal ANGPTL1 attenuates colorectal cancer liver metastasis by regulating Kupffer cell secretion pattern and impeding MMP9 induced vascular leakiness

Kai Jiang et al.

Summary: Exosomal ANGPTL1 attenuated CRC liver metastasis and impeded vascular leakiness in the liver pre-metastatic niche by reprogramming Kupffer cells and decreasing MMP9 expression.

Clinical Implications of Colorectal Cancer Stem Cells in the Age of Single-Cell Omics and Targeted Therapies

Markus H. Frank et al.

Summary: The concept of cancer stem cells (CSCs) suggests a hierarchy within tumors where a minority of more primitive cells give rise to more differentiated cells that make up the tumor bulk, but are not capable of perpetuating tumors on their own. CSCs may drive therapeutic resistance and cancer progression, and their identification and isolation methods have been established. Single-cell omics technologies offer opportunities to discover novel molecular pathways for CRC eradication.
