Development and Validation of an Abridged Version of the REVEAL 2.0 Risk Score Calculator, REVEAL Lite 2, for Use in Patients With Pulmonary Arterial Hypertension

Raymond L. Benza et al.

Summary: REVEAL Lite 2, a simplified version of REVEAL 2.0, provides a convenient method for risk assessment in PAH patients. It includes six noninvasive variables and effectively discriminates among low, intermediate, and high-risk patients for 1-year mortality, showing promise for routine clinical implementation.

ChIP-seq of plasma cell-free nucleosomes identifies gene expression programs of the cells of origin

Ronen Sadeh et al.

Summary: Cell-free DNA in human plasma can provide important molecular information about pathological processes, with cfChIP-seq being a valuable tool to analyze active chromatin modifications in different samples. The study revealed the potential of cfChIP-seq in diagnosing diseases and understanding physiological and pathological processes using blood samples.

Cell-free DNA maps COVID-19 tissue injury and risk of death and can cause tissue injury

Temesgen E. Andargie et al.

Summary: Plasma cfDNA levels were significantly elevated in COVID-19 patients compared to those with influenza or respiratory syncytial virus and healthy controls, with diverse tissue sources including hematopoietic cells, vascular endothelium, hepatocytes, adipocytes, kidney, heart, and lung. cfDNA levels correlated positively with COVID-19 severity, C-reactive protein, and D-dimer, and the cfDNA profile at admission could predict the need for intensive care or death during hospitalization. Increased cfDNA in COVID-19 patients led to excessive mitochondrial ROS production in renal tubular cells, which was inhibited by a TLR9-specific antagonist.

Severe Pulmonary Arterial Hypertension Is Characterized by Increased Neutrophil Elastase and Relative Elafin Deficiency

Andrew J. Sweatt et al.

Summary: The study revealed that PAH patients have elevated levels of NE and decreased levels of elafin, which are associated with more severe clinical disease and outcomes. NE levels can serve as a specific biomarker for the treatment of elafin.
