Summary: Novel cycloalkynes were created by integrating reagent destabilization and transition state stabilization approaches, leading to stable and active SPAAC reagents for bioorthogonal bioconjugation. Computational and experimental studies showed similar SPAAC reactivities for different heterocyclic cycloalkynes. The stability-reactivity balance of strained systems is influenced by the interplay of two electronic effects.
