Summary: Bone marrow is a heterogeneous environment containing various cells, with changes in osteoblast and adipocyte numbers being critical factors in bone marrow adipose tissue (BMAT) production. Targeting the formation of BMAT could be a more effective approach in treating osteoporosis than blocking bone-destroying molecules produced by BMAT. Understanding the molecular basis of increased BM adiposity during pathologic bone loss is essential for developing therapeutic strategies.

Bone targeting antioxidative nano-iron oxide for treating postmenopausal osteoporosis

Summary: This study demonstrates that iron oxide nanoparticles (IONPs) can alleviate estrogen deficiency-induced osteoporosis by regulating local immunity in bone tissues. IONPs scavenge reactive oxygen species and promote osteogenic differentiation while inhibiting osteoclast differentiation. Additionally, the use of alendronate to deliver IONPs to bone tissues enhances therapeutic efficacy. The bone targeting ability, therapeutic efficiency, and biocompatibility of this novel bone targeting iron oxide is verified in ovariectomy-induced osteoporotic mice.
