Quality control and product differentiation of LMWHs marketed in China using 1H NMR spectroscopy and chemometric tools

Summary: In this study, 80 batches of three types of LMWHs marketed in China from different manufacturers were analyzed using 1H NMR experiments and chemometric analysis. The method was able to monitor impurities, contaminants, and batch-to-batch consistency, as well as differentiate biosimilar LMWHs from different manufacturers based on their slightly different structural compositions. Through this method, the quality and safety of LMWHs in the Chinese market were initially assessed.

Chemobiocatalytic Synthesis of a Low-Molecular-Weight Heparin

Summary: Heparin products are widely used as anticoagulants in modern medicine. Low-molecular-weight heparins (LMWHs) are prepared through controlled chemical or enzymatic depolymerization of unfractionated heparins (UFHs) extracted from animal tissues. This study introduces a novel chemoenzymatic process to prepare a chemobiosynthetic LMWH from a low-molecular-weight heparosan, which matches the pharmacopeial specifications for enoxaparin, a LMWH prepared from animal-derived UFHs.

Influence of saccharide modifications on heparin lyase III substrate specificities

Summary: A library of synthetic heparan sulfate oligosaccharides was used to analyze the substrate specificities of heparin lyase III enzymes from two different sources. The study found that specific modifications influenced the digestion of the oligosaccharides, and there were differences in substrate specificities between the two lyase III enzymes for highly sulfated oligosaccharides. These findings provide insights into the structure/function relationships of sulfated domains in biological processes.

The assay used different substrates to determine the enzyme's kinetic parameters and demonstrated the specificity of mNDST1. This study showed that the mNDST1 coupled assay can be used for evaluating enzyme inhibitors for drug development.
