Rapamycin relieves the cataract caused by ablation of Gja8b through stimulating autophagy in zebrafish

Summary: This study revealed that deletion of Gja8b in zebrafish led to severe defects in organelle degradation, a major cause of cataractogenesis in developing lens. The inhibition of autophagy induced deficient organelle degradation, while activation of autophagy using rapamycin relieved the defects, suggesting a potential treatment for cataracts. Furthermore, interaction between GJA8 and ATG proteins was found, and overexpression of GJA8 stimulated autophagy in HLE cells, highlighting a novel cataractogenesis mechanism caused by Gja8b ablation.

Autophagy inhibition attenuates TGF-β2-induced epithelial-mesenchymal transition in lens epithelial cells

Summary: This study demonstrated that TGF-beta 2 induces autophagy flux during EMT progression of LECs, with autophagy activation enhancing fibrogenic responses and cell migration, while autophagy inhibition alleviates EMT markers and cell migration. Additionally, modulation of Smad signaling by autophagy was observed, and ATG7-silenced LECs showed antifibrosis effect induced by TGF-beta 2. Overall, intervention/inhibition of autophagy could attenuate TGF-beta 2-induced EMT in LECs, providing insights for preventing and treating fibrotic cataracts and other fibrotic diseases.
