Human genetic variant E756del in the ion channel PIEZO1 not associated with protection from severe malaria in a large Ghanaian study

Thorsten Thye et al.

Summary: The study did not find evidence to support a strong protective effect of the PIEZO1 E756del variant on severe malaria susceptibility, contradicting previous associations.

Mechanosensitive Piezo1 channels mediate renal fibrosis

Xiaoduo Zhao et al.

Summary: This study reveals the significant role of Piezo1 in kidney fibrosis, where it activates the calcium/calpain2/integrin beta(1) pathway to promote fibrotic responses in renal proximal tubular cells. Moreover, the study demonstrates that inhibition or silencing of Piezo1 effectively prevents fibrotic alterations in these cells.

A role of PIEZO1 in iron metabolism in mice and humans

Shang Ma et al.

Summary: Iron overload can cause progressive organ damage and is associated with genetic factors affecting iron homeostasis. Certain individuals with hereditary disorders may develop iron overload, and some GOF PIEZO1 alleles are linked to iron imbalance. The study demonstrates the key role of PIEZO1 in macrophage phagocytic activity and iron metabolism, with a particular GOF PIEZO1 allele showing a strong association with increased plasma iron levels in individuals of African descent.

Complex Modes of Inheritance in Hereditary Red Blood Cell Disorders: A Case Series Study of 155 Patients

Immacolata Andolfo et al.

Summary: This study analyzed 155 consecutive patients with clinical suspicion of hereditary erythrocyte defects, obtaining an overall diagnostic yield of 84% with monogenic inheritance seen in 69% and multi-locus inheritance in 15% of the tested patients. Some patients showed dual molecular diagnosis of different red blood cell disorders, indicating the importance of genetic testing for patients with complex modes of inheritance.
