A novel concept of Pyridoxal 5′-phosphate permeability in E.coli for modulating the heterologous expression of PLP dependent proteins

Vijay Kumar Saxena et al.

Summary: The study indicated that PLP has a significant impact on the expression and catalytic activity of PLP-dependent proteins in E. coli, with a positive correlation between PLP concentration and expression levels. E. coli does not secrete any dephosphorylation factor for PLP, suggesting internalization of PLP in the cells. The transport mechanism of PLP remains unknown and requires further investigation.

Summary: Gamma-aminobutyric acid (GABA) is a crucial neurotransmitter inhibitor in the mammalian nervous system and can also be used for bioplastics synthesis. Current whole-cell bioconversion systems for GABA production require a costly cofactor PLP. This study introduces a new GABA producing system that can achieve competitive level production without additional PLP supply.

Summary: Transcription factor decoys can effectively regulate gene expression, with tunability through changes in copy number or modifications to the DNA decoy site sequence. Introducing the decoy system can significantly increase arginine production in metabolic flux steering, without affecting growth compared to wild type strains.

Versatile selective evolutionary pressure using synthetic defect in universal metabolism

Lara Selles Vidal et al.

Summary: Designing enzymes with new or improved properties is challenging, and linking the improvement in the target to cell growth through artificial selection pressure is a broadly-applicable approach. Industrial applications often require new or improved enzymes, and a semi-rational approach involving diversification of parent enzymes followed by evaluation of isolated variants can be effective.

Engineering pyridoxal kinase PdxY-integrated Escherichia coli strain and optimization for high-level 5-aminolevulinic acid production

Chengfeng Xue et al.

Summary: By engineering a new strain of Escherichia coli called PIECE, the production of ALA was significantly increased, with more carbon being redirected into the TCA cycle to generate more glycine and glutamate, resulting in increased ALA yield and biomass. The results suggest that PIECE not only allows for high-level ALA production but also has potential to produce value-added chemicals in the future by reprogramming carbon flux.
