Three-dimensional hierarchical plasmonic nano-architecture based label-free surface-enhanced Raman spectroscopy detection of urinary exosomal miRNA for clinical diagnosis of prostate cancer

Woo Hyun Kim et al.

Summary: Urinary exosomal miRNAs have emerged as valuable biomarkers for early-stage diagnosis and prognosis of prostate cancer due to their non-invasiveness, stability, and representation of cell status. However, developing an accurate detection method for urinary exosomal miRNAs is challenging due to their low abundance and sequence homology. In this study, a label-free miRNA sensing platform using surface-enhanced Raman scattering (SERS) was developed, based on a three-dimensional hierarchical plasmonic nano-architecture, for the detection of urinary exosomal miRNAs. Clinical validation using urinary samples demonstrated that the SERS sensor achieved high diagnostic accuracy for distinguishing prostate cancer patients from healthy controls based on differential expression of urinary exosomal miRNAs. This study highlights the potential of the SERS sensor for facile and accurate measurement of miRNA expression and its importance in disease diagnosis.
