Direct observation of chemo-mechanical coupling in DnaK by single-molecule force experiments

Summary: Protein allostery requires a communication channel for functional regulation between distal sites within a protein. In the molecular chaperone Hsp70, the ATP/ADP status of a nucleotide-binding domain regulates the substrate affinity of a substrate-binding domain. Using laser optical tweezers, the mechanical properties of a truncated two-domain DnaK were examined in different states. It was found that in the ATP state, the substrate-binding domain becomes mechanically destabilized due to structural changes.

Summary: Researchers characterized biochemical properties of selected domain-interface mutants in bacterial Hsp70 DnaK and developed a theoretical model to explain allosteric equilibria among Hsp70 conformational states. They proposed a restraining state and a stimulating state to describe the different characteristics of Hsp70 in ATP hydrolysis and peptide binding.
