APR-246 induces early cell death by ferroptosis in acute myeloid leukemia

Summary: APR 246 is a promising therapeutic agent for myelodysplastic syndromes and acute myeloid leukemia that targets p53 mutated proteins. It has been found to reactivate the transcriptional activity of p53 mutants and induce p53-independent cell death. Summary: A new series of benzodiazepines and chiral Schiff bases were prepared by in situ condensation of (S)-(-)-perillaldehyde with various amines, hydrazines, and oxalyl dihydrazide. The enantiomeric compounds TK16 were obtained in moderate to good yields after spontaneous precipitation in ethanol, and their structures were identified through NMR experiments, FTIR, and single-crystal X-ray. The new derivatives showed good antioxidant and antibacterial activities after screening.

Summary: Chemotherapeutic agents, including the cardiac glycoside oleandrin, have been shown to induce immunogenic cell death (ICD) in breast cancer cells, enhancing anti-tumor immune responses. Oleandrin treatment triggered ICD in breast cancer cells by inducing calreticulin exposure and release of HMGB1, HSP70/90, and ATP. Mechanistically, oleandrin induced ER stress-associated, caspase-independent ICD mainly through the PERK/elF2 alpha/ATF4/CHOP pathway, suggesting its potential in combination with immune checkpoint inhibitors to improve immunotherapy efficacy.
