Autophagy as a molecular target of quercetin underlying its protective effects in human diseases

Milad Ashrafizadeh et al.

Summary: Autophagy is a self-eating process that degrades aged or damaged components and organelles. It plays a crucial role in physiological conditions and neurological disorders. Quercetin, a plant-derived chemical, has various biological and therapeutic effects. This review provides an overview of the relationship between quercetin and autophagy, as well as how quercetin modulates the autophagy mechanism.

Quercetin and vitamin E ameliorate cardio-apoptotic risks in diabetic rats

Oluwatoyin O. Ojo et al.

Summary: Apoptosis is increased in diabetes and mitochondria play a crucial role in the pathophysiology of diabetes. Quercetin and vitamin E have shown potential in delaying diabetes-induced complications, potentially by regulating the opening of the mitochondrial permeability transition (mPT) pore. The study found that quercetin and vitamin E treatment reversed the increased swelling and lipid peroxidation in diabetic hearts, suggesting their anti-apoptotic effect by inhibiting mPT pore and modulating mitochondrial-mediated apoptosis.

Relevance of mitochondrial oxidative stress to arrhythmias: Innovative concepts to target treatments

Can Liu et al.

Summary: Cardiac arrhythmia is a common and potentially fatal condition worldwide. Mitochondrial oxidative stress plays a crucial role in its development, affecting the normal function of cardiomyocytes through ion channels and related proteins. Targeted treatments focusing on mitochondria may offer new possibilities for managing arrhythmia by reducing its progression and occurrence of severe episodes, and could be effective in personalized disease prevention.

Cardiomyocyte Na+ and Ca2+ mishandling drives vicious cycle involving CaMKII, ROS, and ryanodine receptors

Bence Hegyi et al.

Summary: Cardiomyocyte mishandling of Na+ and Ca2+, upregulated CaMKII, and increased ROS are characteristics of heart diseases. Inhibition of CaMKII, I-NaL, and RyRs can attenuate APD prolongation and STV increase, with potential therapeutic effects against arrhythmias and diseases. Targeting the Na+-Ca2+-CaMKII-ROS-I-NaL vicious cycle may provide novel integrated insight for treating cardiac arrhythmias and diseases.

Cytoprotective effects of the flavonoid quercetin by activating mitochondrial BKCa channels in endothelial cells

Rafal Pawel Kampa et al.

Summary: The natural flavonoid quercetin acts as a mitoBKCa channel opener, increasing channel open probability and protecting cells from damage. It enhances cellular migration and depolarizes the mitochondrial membrane, while also increasing gene expression and protein levels of mitoBKCa 133 regulatory subunit. The cytoprotective effects suggest the involvement of BKCa channel activation, providing a new mitochondrial pathway beneficial for vascular endothelial cells.

Baode Shen et al.

Summary: The study revealed that the size of the nanoparticles affects the absorption and biodistribution of the drug, with smaller particles being more easily absorbed but dissolving faster in vivo, leading to higher drug distribution but lower accumulation of integral nanocrystals in the liver. Larger particles, on the other hand, show slower dissolution and higher liver distribution due to enhanced recognition by RES. The absolute exposure of integral nanocrystals to oral bioavailability enhancement was unexpectedly high, indicating a significant difference in absorption and biodistribution between integral nanocrystals and overall drugs following oral and intravenous administration.
