Hybrid spheroid microscaffolds as modular tissue units to build macro-tissue assemblies for tissue engineering ✩

Olivier Guillaume et al.

Summary: Tissue engineering and regenerative medicine (TERM) has previously relied on scaffold-based or scaffold-free strategies, but a new approach called the third TERM strategy, involving highly porous microscaffolds and single spheroids, has the potential to overcome limitations of previous options. A systematic study comparing microscaffold-based tissue units with conventional spheroids demonstrated that the microscaffold did not impede spheroid formation or impact viability. Additionally, the fusion and differentiation potential necessary for TERM applications were preserved. Microscaffold-based tissue units offer benefits such as enhanced cell retention, decreased compaction, and better size control for tissue constructs formed through self-assembly.

miRNA induced 3D bioprinted-heterotypic osteochondral interface

Nazmiye Celik et al.

Summary: This study presents a novel approach to osteochondral reconstruction using 3D bioprinting of miR-transfected ADSC spheroids. By transfecting specific miRNAs, spheroids expressed genes and protein markers related to osteogenic and chondrogenic differentiation, leading to enhanced mineralization and cell proliferation. Through aspiration-assisted bioprinting, these spheroids were printed into a stratified interface with distinct osteogenic and chondrogenic zones. This approach holds great promise for the biofabrication of various tissue interfaces, not limited to osteochondral interfaces.
