In vivo imaging of calcium dynamics in zebrafish hepatocytes

Macarena Pozo-Morales et al.

Summary: This study successfully observed the changes in hepatocyte calcium dynamics by taking advantage of the transparency of zebrafish larvae. The presence of calcium oscillations, waves, and spikes in vivo were demonstrated, and it was found that calcium waves are related to nutrition intake and negatively regulate starvation-induced lipid droplet accumulation.

Towards an integrative understanding of cancer mechanobiology: calcium, YAP, and microRNA under biophysical forces

Chenyu Liang et al.

Summary: This paper reviews the interplay between microenvironmental mechanics and biochemical-genetic activities in tumor progression. The authors focus on the roles of calcium ions, Yes-associated protein, and microRNAs in this process. They propose future directions for cancer mechanobiology research using these effectors.

Hydrolytically Degradable Microgels with Tunable Mechanical Properties Modulate the Host Immune Response

Maria M. Coronel et al.

Summary: This study demonstrates a facile method to synthesize microgels crosslinked with ester-containing junctions and investigates the effect of microgel degradability on in vivo tissue responses. The injectable microgels show time-dependent degradation in vivo and have the potential to modulate host tissue immune responses.

A single-cell analysis of breast cancer cell lines to study tumour heterogeneity and drug response

G. Gambardella et al.

Summary: This study characterizes the cell lines of different subtypes of breast cancer using single-cell RNA-seq and trains a deconvolution algorithm to predict drug responses in heterogeneous tumor cell populations. The findings show high heterogeneity in the expression of biomarkers within cancer cells, and transcriptional heterogeneity allows cells with differential drug sensitivity to coexist in the same population. This work provides a framework to determine tumor heterogeneity in terms of cell line composition and drug response.

Molecular pharmacology of P2Y receptor subtypes

Ivar von Kuegelgen

Summary: P2Y receptors, divided into different subtypes, play important roles in various physiological and pathological processes, including pain and inflammation. Antagonism of platelet P2Y12 receptors and activation of P2Y2 receptors have been utilized in treating thrombotic complications and dry eye syndrome, respectively, demonstrating their potential therapeutic importance. Structural information obtained through crystallography and molecular modeling will aid in the design of novel selective drugs targeting these receptors.

The genie in the bottle-magnified calcium signaling in dorsolateral prefrontal cortex

Amy F. T. Arnsten et al.

Summary: Neurons in association cortices are vulnerable in cognitive disorders, with molecular mechanisms for higher cognitive operations leading to dysfunction and neurodegeneration when regulation is lost. Excessive cAMP-calcium signaling can weaken synaptic efficacy and drive neurodegeneration, necessitating tight regulation to prevent cognitive disorders.

Mechanisms Underlying Connexin Hemichannel Activation in Disease

Raf Van Campenhout et al.

Summary: Gap junctions and connexin hemichannels play different roles in intercellular and extracellular communication, with the former maintaining homeostasis and the latter being associated with the onset and spread of disease by mediating messenger transport that fuels inflammation and cell death in various diseases. The present review discusses the mechanisms involved in the activation of connexin hemichannels during pathology.

Activation of Cx43 Hemichannels Induces the Generation of Ca2+ Oscillations in White Adipocytes and Stimulates Lipolysis

Egor A. Turovsky et al.

Summary: The study investigated the mechanisms of Ca2+ oscillation generation and regulation of the lipolysis/lipogenesis balance in white adipocytes in response to a decrease in extracellular calcium concentration. It was found that activation of connexin-43 led to ATP release from adipocytes, causing changes in gene expression related to lipolysis and a decrease in lipid droplet accumulation.

Mechanical and chemical activation of GPR68 probed with a genetically encoded fluorescent reporter

Alper D. Ozkan et al.

Summary: The study developed a genetically encoded fluorescent reporter, iGlow, by inserting a fluorescent protein into the third intracellular loop of GPR68, which responds to physiological GPR68 activators and synthetic agonists with specificity. iGlow activation is unaffected by pharmacological modulation of downstream G-protein signaling or disruption of actin filaments, providing a new tool for investigating GPR68-dependent signaling in health and disease.
