ER-Targeting Cyanine Dye as an NIR Photoinducer to Efficiently Trigger Photoimmunogenic Cancer Cell Death

He Ma et al.

Summary: This study developed the first ER-localizable immunogenic cell death (ICD) photoinducer that induces ER stress under mild near-infrared (NIR) irradiation using a thio-pentamethine cyanine dye (TCyS). The photoinducer has superior ROS generation capability in both normoxia and hypoxia conditions and leads to the production of damage-associated molecular patterns. In vivo experiments demonstrated its effectiveness in tumor treatment.

Emerging photodynamic nanotherapeutics for inducing immunogenic cell death and potentiating cancer immunotherapy

Shenwu Zhang et al.

Summary: Immunotherapy is a promising cancer treatment, but its efficacy for immune-cold tumors is not satisfactory. Photodynamic therapy can effectively induce immunogenic cell death, and the use of nanotechnology enhances the effectiveness of cancer immunotherapy. This article reviews recent advances in photodynamic therapy, nanotechnology, and immunogenic cell death, and discusses the prospects and challenges in this field.

Ultrasound-Augmented Mitochondrial Calcium Ion Overload by Calcium Nanomodulator to Induce Immunogenic Cell Death

Pan Zheng et al.

Summary: This study developed an acid-sensitive nanoparticle ((CaCUR)-Ca-PEG) that induces mitochondrial Ca2+ overload and acts as an ICD inducer in improved cancer therapy. Combination with ultrasound (US) enhances the ICD effect, showing promising therapeutic and diagnostic properties.

Nanocatalytic Innate Immunity Activation by Mitochondrial DNA Oxidative Damage for Tumor-Specific Therapy

Han Jiang et al.

Summary: A novel tumor immunotherapy modality based on nanocatalytic innate immunity activation by inducing oxidative damage in tumor cells' mtDNA is proposed. This approach reactivates macrophages' immune response against cancer cells, leading to the inhibition of primary and distant tumor growth during treatment.
